Biotransformation of Benzaldehyde to L-Phenylacetylcarbinol (L-PAC) by Free Cells of Torulaspora delbrueckii in presence of Beta-Cyclodextrin BABT
Shukla,Vilas. B.; Kulkarni,Pushpa R..
Studies were carried out to explore the possibility of decreasing the toxic and inhibitory effects of the substrate benzaldehyde during its biotransformation to L-PAC by free cells of Torulaspora delbrueckii using beta -cyclodextrin (beta -CD). Use of various levels of benzaldehyde and acetaldehyde in presence of 2% of beta -CD showed that, in presence of beta -CD, the organism could tolerate higher levels of benzaldehyde and acetaldehyde. Semi-continuous feeding of benzaldehyde and acetaldehyde was found to increase the yield of L-PAC in comparison with one time feeding.

Economic model for obtaining cyclodextrins from commercial cgtase BJPS
Koga,Luciana Numata; Fenelon,Vanderson Carvalho; Miyoshi,Juliana Harumi; Moriwaki,Cristiane; Wessel,Kamila Byanca Baldin; Mangolim,Camila Sampaio; Matioli,Graciette.
A repetitive batch process was employed followed by membrane ultrafiltration system to produce low-cost cyclodextrins (CDs) using commercial enzymes Toruzyme® cyclomaltodextrin glucanotransferase (CGTase) and its kinetic parameters were determined. The ultrafiltration system enabled the removalof inhibitory products from the reaction medium, allowing the enzyme to be recovered for reuse. A 10 kDa membrane was used to separate the different CDs produced by the CGTase. The substrates evaluated were maltodextrin, corn starch and cassava starch at 5, 10 and 15% (w/V), in the presence and absence of 10% (V/V) ethanol. After reaction for 132 h, 10% (w/V) cassava starch in the presence of ethanol provided the best results with 32.1 mg/mL of β-CD. Maximum...

Formulation and in vitro evaluation of taste-masked oro-dispersible dosage form of diltiazem hydrochloride BJPS
Jagdale,Swati Changdeo; Gawali,Vaibhav Uttam; Kuchekar,Bhanudas Shankar; Chabukswar,Aniruddha Rajaram.
Diltiazem hydrochloride is a calcium channel blocker generally indicated for the treatment of angina and hypertension, and it is extensively metabolized due to the hepatic metabolism. Formulation of diltiazem hydrochloride into an oro-dispersible dosage form can provide fast relief with higher bioavailability. The bitter taste of the drug should be masked to formulate it in a palatable form. In the present work, an attempt was made to mask the taste by complexation technique, with a formulation into an oro-dispersible dosage form, using superdisintegrants Doshion P544, crospovidone (CP) and sodium starch glycolate (SSG). The complexes of diltiazem hydrochloride with β-CD (1:1 molar ratio) were prepared by kneading, co-evaporation, co-grounding,...

Investigation of aromatic hydrocarbon inclusion into cyclodextrins by Raman spectroscopy and thermal analysis ArchiMer
Tijunelyte, Inga; Dupont, Nathalie; Milosevic, Irena; Barbey, Carole; Rinnert, Emmanuel; Lidgi-guigui, Nathalie; Guenin, Erwann; De La Chapelle, Marc Lamy.
Among various cavitand molecules, cyclodextrins are extensively studied due to their ability to form host-guest complexes with small hydrophobic molecules. Aiming to explore cyclodextrin implementation on the scopes related to the environmental pollution monitoring or remediation, extensive studies for understanding the cyclodextrin-based host-guest complex formation with selected targeted substances are conducted. In this context, two polycyclic aromatic hydrocarbons, naphthalene and fluoranthene as well as toluene as a member of volatile organic compounds, were studied regarding their ability to encapsulate into cyclodextrin cavities. Synthesised complexes were examined by thermogravimetric analysis combined with Raman spectroscopy. The obtained results...

Is there still room to explore cyclodextrin glycosyltransferase-producers in Brazilian biodiversity? Anais da ABC (AABC)
GOMES,ANDREA C.S.M; SANTOS,SAMUEL R. DOS; RIBEIRO,MAYCON C.; CRAVO,PEDRO V.L.; VIEIRA,JOSÉ DANIEL G.; SOUZA,KEILI M.C. DE; AMARAL,ANDRE C..
ABSTRACT In the present work, different Brazilian biomes aiming to identify and select cyclodextrin glycosyltransferase-producer bacteria are explored. This enzyme is responsible for converting starch to cyclodextrin, which are interesting molecules to carry other substances of economic interest applied by textile, pharmaceutical, food, and other industries. Based on the enzymatic index, 12 bacteria were selected and evaluated, considering their capacity to produce the enzyme in culture media containing different starch sources. It was observed that the highest yields were presented by the bacteria when grown in cornstarch. These bacteria were also characterized by sequencing of the 16S rRNA region and were classified as Bacillus, Paenibacillus,...

Targeting of temozolomide using magnetic nanobeads: an in vitro study BJPS
Gürten,Berna; Yenigül,Elçin; Sezer,Ali Demir; Altan,Cem; Malta,Seyda.
Temozolomide, a chemotherapeutic drug that is often administered for the treatment of brain cancer has severe side effects and a poor aqueous solubility. In order to decrease the detrimental effect of the drug over healthy cells, a novel drug delivery vehicle was developed where the therapeutic drug was encapsulated within the hydrophobic cavities of b-CD modified magnetite nanoparticles, which are embedded in chitosan nanobeads prepared by salt addition. In-vitro studies have shown that the magnetic properties of the novel delivery vehicle are adequate for targeted drug delivery applications under an external magnetic field. Additionally, an increase in the amount of chitosan was shown to exhibit a strong shielding effect over the magnetic properties of...
